Gram staining
Dye solutions:
Solution A: Carbol gentian violet/crystal violet
Solution B: Lugol’s solution
Solution C: Ethanol (95%, methylated spirits)
Solution D: Safranin
Method:
1. Production of dry specimen
2. Allow Solution A to act for 90 seconds
3. Rinse with water
4. Allow Solution B to act for 60 seconds
5. Rinse with water
6. Sluice with ethanol (Solution C)
7. Rinse with water
8. Allow Solution D to act for 60 seconds
9. Rinse with water
10. Dry
